Comparison of Dumas and Kjeldahl methods for determination of nitrogen in feces.

چکیده
The present study was undertaken to compare the nitrogen content of human feces as determined by the Kjeldahl technic and the micro-Dumas method as utilized in the automated Coleman nitrogen analyzer. The standard deviation between duplicate determinations of the same sample was 0.06% nitrogen in the case of the nitrogen analyzer and 0.02% nitrogen for the Kjeldahl method. The standard deviation between the average values obtained by each method for each sample was 0.07% nitrogen. The agreement between the two methods seemed to be independent of the nitrogen content. The apparent recovery of nitrogen by the nitrogen analyzer was found to be greater than that recovered by the Kjeldahl procedure. Factors which may be involved in this difference in recovery between the two methodsare discussed.

Total nitrogen determination
As a preliminary to the consideration of automated, or more correctly mechanized, methods for the determination of total organic nitrogen the discussion of some general aspects of nitrogen determination is illuminating. There are at least four recent reviews, for example those by Bradstreet (1965), Fleck & Munro (I965), Jacobs (1965), and Munro & Fleck (1969), of the determination of nitrogen f...
متن کاملThe Jean Dumas Method
Jean Dumas (Fig. 1) published his combustion method in Paris back in 1848. This makes the method considerably older than Kjeldahl digestion. While researching the composition of natural oils, Dumas tried combusting his samples, initially to analyse the carbon and hydrogen contained therein. Combustion therefore makes it possible to analyse multiple parameters, while Kjeldahl digestion is only s...
متن کاملAn absolute method for protein determination based on difference in absorbance at 235 and 280 nm.
Because of the importance of quantitative determination of protein in the research laboratory as well as in the food and feed industries (I), search for the ideal method continues unabated after many years. Methods available include nitrogen determination (Kjeldahl (2) and Dumas (3)), hydrolysis of the protein, derivatization of the amino acids with phthalaldehyde and fluorescence determination...
متن کاملKjeldahl nitrogen analysis as a reference method for protein determination in dairy products.
Measurement of total nitrogen by Kjeldahl analysis is the historical reference method for determination of the protein content of dairy products and is used for both calibration and validation of alternative methods for protein determination. Accurate evaluation of alternative methods is not possible if there is large uncertainty regarding the reference values. When Kjeldahl analysis is used to...
متن کاملAdaptation of the Micro-kjeldahl Method for the Determination of Nitrogen in Plant Tissues.
The micro-Kjeldahl method for nitrogen determination has been widely used in organic chemistry laboratories in recent years. The value of this method, developed through the brilliant researches of PiLcH, BANG, PARNAS and WAGNER, PREGL, and coworkers, has justified its application to industrial and clinical problems. Plant physiologists have not generally adopted quantitative micro-methods for n...
